Abstract

Drug resistance can be said to be one of the main causes of treatment failure in cancer. Through various mechanisms, malignant cells rapidly develop resistance to chemotherapeutic agents. Additionally, the problem of drug resistance appears to have affected the quest to develop new anticancer drugs. To improve chemotherapy and identify specific targets for new drugs, it is imperative to understand the various mechanisms of drug resistance. This chapter gives an insight into the pharmacokinetic, pharmacodynamic and pharmacogenomic factors that cause drug resistance in chemotherapy. Furthermore, factors to consider in discovery and development of new chemotherapeutic agents are also discussed.
